Biological Background: BiP/GRP78 Function and Localization
- Endoplasmic reticulum chaperone BiP, also known as GRP-78, Binding-immunoglobulin protein, and HSPA5, is a member of the heat shock protein 70 (HSP70) family encoded by the HSPA5 gene.
- Functions as a key ER chaperone involved in protein folding and quality control, facilitating correct folding and degradation of misfolded proteins through interaction with DNAJC10/ERdj5. Related references: PMID:2294010, PMID:23769672, PMID:23990668
- Acts as a repressor of the unfolded protein response (UPR) by binding and inactivating EIF2AK3/PERK and ERN1/IRE1 under unstressed conditions; accumulation of misfolded proteins releases BiP, allowing UPR activation. Related references: PMID:11907036, PMID:1550958, PMID:19538957
- Plays an auxiliary role in post-translational transport of small presecretory proteins across the ER membrane, likely cooperating with SEC62 to modulate the SEC61 translocon channel, and may be involved in apoptosis and cell proliferation. Related references: PMID:26045166
- Serves as a cellular receptor for several flaviviruses, including Dengue virus, Zika virus, and Japanese encephalitis virus, mediating their binding and entry into host cells. Related references: PMID:15098107, PMID:28053106, PMID:33432092
- Acts as a receptor for CotH proteins expressed by mucorales fungi, facilitating epithelial cell invasion in mucormycosis; specifically interacts with R.delemar CotH3 in nasal epithelial cells during rhinoorbital/cerebral mucormycosis. Related references: PMID:20484814, PMID:24355926, PMID:32487760
- Localizes primarily to the endoplasmic reticulum lumen, but is also detected in melanosomes, cytoplasm, and at the cell surface, reflecting its diverse functional roles.
- Subject to numerous post-translational modifications including acetylation, phosphorylation, methylation, nitration, and ubiquitination, which may regulate its chaperone activity and interactions.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide derived from human BiP/GRP78; due to high sequence homology, cross-reactivity with mouse and rat is expected but should be validated in your target species.
- For Western blot, a band around 72 kDa is predicted, though post-translational modifications or experimental conditions may cause slight shifts; include appropriate positive controls and molecular weight markers.
- For IHC-P and IF/ICC, optimize antigen retrieval and permeabilization steps to detect ER-localized BiP; consider using ER-specific counterstains to confirm subcellular localization.
- In ELISA applications, ensure proper coating conditions and test serial dilutions of the antibody to establish optimal working concentrations for your specific assay format.
