Biological Background: BIN2 (Shaggy-related protein kinase eta) Function and Localization

  • BIN2 is a shaggy-related protein kinase eta (ASK-eta), also known as protein BRASSINOSTEROID INSENSITIVE 2 and protein ULTRACURVATA 1. It belongs to the shaggy-like kinase family in Arabidopsis thaliana.
  • Functions as a central negative regulator in the brassinosteroid signal transduction pathway, essential for normal plant growth and development. It may also be involved in auxin signaling.
  • Phosphorylates transcription factors BZR1 and BZR2/BES1, promoting their proteasomal degradation to dampen brassinosteroid-responsive gene expression.
  • Upon brassinosteroid perception, inhibits stomatal development by phosphorylating and inactivating the MAPKK kinase YDA and MAPK kinases MKK4/MKK5. Related references: PMID:11847343 PMID:12114546 PMID:12427989
  • Phosphorylates BR-signaling kinases BSK1, 3, 5, 6, 8, and 11 in vitro, further modulating signal transduction. Related references: PMID:23496207
  • Phosphorylates and destabilizes WRKY transcription factors WRKY46, WRKY54, and WRKY70, linking brassinosteroid signaling to plant immunity. Related references: PMID:28576847
  • Plays a critical role in stomatal development by mediating BASL nuclear exclusion and promoting asymmetric cell division. It phosphorylates and inhibits SPCH in the nucleus of stomatal lineage ground cells, restricting cell division and promoting differentiation. Related references: PMID:22466366 PMID:30429609
  • Localizes to multiple subcellular compartments including the cytoplasm, cell cortex, nucleus, and cell membrane, allowing versatile signaling functions.
  • Exhibits tissue-specific expression in developing seed coat, suspensor, and stomatal lineage cells. Related references: PMID:10080716 PMID:30429609

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide covering amino acids 281–380 of BIN2. This region may represent a conserved functional domain; consider verifying its relevance to your experimental context.
  • For Western blot, a band at approximately 43 kDa is expected. Use whole-cell lysates from Arabidopsis thaliana tissues and optimize blocking and detection conditions.
  • The antibody is suitable for ELISA; validate coating conditions and include proper positive/negative controls.
  • Expected to recognize Arabidopsis thaliana BIN2. If applying to other species or related kinases, determine cross-reactivity experimentally.
  • Shipped on ice bags; store appropriately upon arrival and avoid repeated freeze-thaw cycles.
