Biological Background: Bif-1 Function and Localization
- Endophilin-B1 (Bif-1, SH3GLB1) is a 41 kDa protein encoded by the SH3GLB1 gene in humans, existing as two alternatively spliced isoforms with distinct roles: isoform 1 promotes apoptosis, while isoform 2 protects neuronal viability.
- Bif-1 plays a critical role in mitochondrial outer membrane dynamics and is required for normal mitochondrial morphology and function.
Related references: PMID:15452144 - It is a key mediator of caspase-dependent apoptosis by promoting BAX/BAK1 activation and is also involved in caspase-independent cell death during nutrient starvation.
Related references: PMID:16227588 - Bif-1 activates the lipid kinase PIK3C3 (VPS34) during autophagy by associating with the PI3K complex II (PI3KC3-C2), regulating autophagosome formation and ATG9A trafficking from the Golgi.
Related references: PMID:17891140 PMID:21068542 - It participates in membrane remodeling events, including Golgi membrane tubulation and fragmentation, and may promote membrane fusion by recruiting proteins to high-curvature membranes.
Related references: PMID:11604418 - Subcellularly, Bif-1 localizes to the cytoplasm, Golgi apparatus, mitochondrial outer membrane, autophagosome membrane, and the midbody during cytokinesis.
- Tissue expression is highest in heart, skeletal muscle, kidney, and placenta, with lower levels in brain, colon, thymus, spleen, liver, small intestine, lung, and peripheral blood leukocytes.
- Post-translationally, Bif-1 is subject to acetylation and phosphorylation; it features an SH3 domain and coiled-coil regions critical for protein interactions and membrane association.
Experimental Guidance and Technical Tips
- For IF/ICC, use appropriate fixation (e.g., 4% PFA) and permeabilization (e.g., 0.1% Triton X-100). Consider including a blocking step with 5% BSA to reduce background.
- In ELISA, perform a dilution series (e.g., starting from 1:500) to determine the optimal antibody concentration for your specific antigen coating and detection system.
- Validate antibody specificity in human cell lines known to express Bif-1, such as HeLa or HEK293. Include a negative control (e.g., siRNA knockdown) to confirm target recognition.
- Since this is a polyclonal antibody, be aware of potential batch-to-batch variability; we recommend qualifying each new lot in your assay.
