Biological Background: Bif-1 Function and Localization
- Endophilin-B1 (Bif-1, also known as CGI-61, PPP1R70) is an SH3 domain-containing member of the endophilin family, characterized by an N-terminal BAR domain and a C-terminal SH3 domain.
- Plays a role in mitochondrial dynamics, promoting normal outer mitochondrial membrane morphology and function. Related references: PMID:15452144
- May recruit proteins to highly curved membranes and facilitate membrane fusion events. Related references: PMID:11604418
- Promotes caspase-dependent apoptosis by enhancing BAX/BAK1 activation; isoform 1 is proapoptotic, while isoform 2 acts antiapoptotically in neuronal cells and supports mitochondrial morphology maintenance. Related references: PMID:16227588
- Activates PIK3C3 lipid kinase activity within the PI3K complex II during autophagy, and regulates ATG9A trafficking from the Golgi to peripheral cytoplasm for autophagosome formation via Golgi membrane tubulation. Related references: PMID:17891140 PMID:21068542
- Participates in degradative endocytic trafficking and cytokinesis, likely as part of the PI3KC3-C2 complex. Related references: PMID:20643123
- Localizes to the cytoplasm, Golgi apparatus membrane, mitochondrial outer membrane, cytoplasmic vesicles, autophagosome membrane, and midbody.
- Highly expressed in heart, skeletal muscle, kidney, and placenta; also detected at lower levels in brain, colon, thymus, spleen, liver, small intestine, lung, and peripheral blood leukocytes.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region (amino acids 120-365) of human Bif-1 that is well conserved across human, mouse, and rat, which supports the observed cross-reactivity. Verify performance in the species of interest.
- For Western blotting, consider using whole cell lysates from tissues with high endogenous expression, such as heart or skeletal muscle. A band near 41 kDa is expected.
- For immunohistochemistry (paraffin), appropriate antigen retrieval may be required. Validate staining patterns against known localization in tissue sections.
- For ELISA applications, the antibody may be used as capture or detection reagent depending on assay design. Optimization of working concentrations is recommended.
