Biological Background: BICD2 Function and Localization

  • BICD2 (Protein bicaudal D homolog 2), also referred to as SMALED2, is encoded by the BICD2 gene (Gene ID 23299).
  • Functions as an adaptor protein that links the dynein motor complex to various cargos and converts dynein into a highly processive motor in the presence of dynactin. Related references: PMID:25814576
  • Facilitates the binding of RAB6A to the Golgi and regulates COPI-independent Golgi-to-ER transport by recruiting the dynein-dynactin motor complex. Related references: PMID:25962623
  • Contributes to proper nuclear and centrosomal positioning before mitosis by regulating dynein and kinesin-1; during G2, associates with RANBP2 at nuclear pores to recruit dynein.
  • Subcellularly localized at the Golgi apparatus, cytoplasm, cytoskeleton, nuclear envelope, and nuclear pore complex.
  • Ubiquitously expressed across tissues.
  • Post-translational modifications include acetylation and phosphorylation; linked to neurodegenerative disease variants (keywords: mRNA transport, neurodegeneration, protein transport, translocation, transport).

Experimental Guidance and Technical Tips

  • The immunogen was designed using a recombinant protein corresponding to amino acids 440–620 near the C-terminus of human BICD2. For Western blot, a band at approximately 94 kDa is expected; validate in the relevant sample system (e.g., human, mouse, or rat cell lysates).
  • For ELISA, the polyclonal nature may provide robust detection; consider sandwich ELISA configurations with a matched antibody pair if quantitation is needed.
  • Cross-reactivity with human, mouse, and rat has been reported; however, verify performance in your specific tissue or cell line of interest.
