Biological Background: BHLHE41 Function and Localization

  • BHLHE41 (also known as DEC2, SHARP1, bHLHb3) is a basic helix-loop-helix transcription factor that functions as a transcriptional repressor, primarily involved in the regulation of circadian rhythms by negatively controlling clock gene expression.   • It forms a negative autoregulatory feedback loop (DEC loop), distinct from the PER/CRY loop, and represses its own expression as well as that of DBP and PER1; both loops interlock to maintain circadian oscillations. Related references: PMID:11278948, PMID:14672706, PMID:15193144

  • BHLHE41 represses the transcriptional activity of the CLOCK-BMAL1 heterodimer by competing for E-box elements (5'-CACGTG-3') in target gene promoters, thereby attenuating circadian output. Related references: PMID:25083013
