Biological Background: BHLHE40 Function and Localization

  • BHLHE40 is a Class E basic helix-loop-helix transcriptional repressor that plays a critical role in circadian rhythm regulation by negatively regulating clock genes and clock-controlled genes. Related references: PMID:12397359, PMID:18411297
  • It acts as the negative limb of an autoregulatory feedback loop (DEC loop) distinct from the PER/CRY loop, repressing PER1/2 and being itself repressed by PER1/2 and CRY1/2. Related references: PMID:14672706, PMID:15193144
  • Represses circadian transcriptional activators by competing with CLOCK-BMAL1 heterodimers for E-box binding in target gene promoters. Related references: PMID:15560782
  • Negatively regulates its own expression as well as DBP and BHLHE41/DEC2. Related references: PMID:14672706
  • Functions as a corepressor for RXR and RXR-LXR heterodimers, inhibiting ligand-induced transactivation of RXRA and NR1H3/LXRA. Related references: PMID:19786558
  • Involved in chondrocyte differentiation via the cAMP pathway and contributes to circadian blood pressure control by repressing ATP1B1 transcription. Related references: PMID:19786558, PMID:30012868
  • Subcellular localization includes both cytoplasm and nucleus, consistent with its transcriptional regulatory role.
  • Broadly expressed in tissues including cartilage, spleen, intestine, lung, heart, brain, liver, muscle, and stomach.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 121–300, a region that may include functional domains such as the bHLH domain and the Orange domain; consider verifying epitope specificity with truncated constructs if needed.
  • For western blot, the predicted molecular weight is ~46 kDa; however, post-translational modifications may cause a shift, so validate using positive control lysates from tissues like cartilage or spleen.
  • For immunohistochemistry (IHC-P), antigen retrieval conditions should be optimized; test on known expressing tissues and include a nuclear staining control.
  • Cross-reactivity with mouse and rat suggests suitability for these species, but always confirm with appropriate controls in the relevant sample system.
