Biological Background: beta 2 Microglobulin Function and Localization
- Beta-2-microglobulin (B2M) is the light chain component of the class I major histocompatibility complex (MHC), essential for the presentation of peptide antigens to the immune system.
- Exogenously applied M. tuberculosis EsxA or EsxA-EsxB (or host-expressed EsxA) binds B2M and reduces its cell surface export, potentially impairing class I antigen presentation. Related references: PMID:25356553
- B2M belongs to the immunoglobulin superfamily and features a classical immunoglobulin domain structure.
- The protein is localized to the cell surface as part of MHC I complexes, and it is also secreted.
- Post-translational modifications include glycation, glycosylation, a conserved disulfide bond, and N-terminal pyrrolidone carboxylic acid formation.
- B2M is associated with amyloidosis (amyloidogenic forms) and has been linked to disease variants.
- It is a key molecule in adaptive immunity, serving as a reference proteome member identified through proteomics.
Experimental Guidance and Technical Tips
- The immunogen consists of a recombinant fusion protein corresponding to amino acids 21-119 of human beta 2 Microglobulin (NP_004039.1). This region includes the mature protein sequence, excluding the signal peptide, and is expected to generate antibodies recognizing the full-length native protein.
- For Western blot (WB), the predicted molecular weight is ~14 kDa. Ensure proper protein loading controls and membrane blocking to minimize background.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval may be necessary; optimization of fixation and permeabilization conditions is recommended.
- Given its polyclonal nature, lot-to-lot consistency should be verified for critical assays; screening by ELISA or dot blot is advisable.
- Cross-reactivity with mouse and rat orthologs is reported, but sequence alignment and pilot experiments are recommended to confirm binding in these species.
