Biological Background: Beta-2-microglobulin Function and Localization
- Beta-2-microglobulin (B2M, also known as IMD43) is the light chain of the major histocompatibility complex class I (MHC I) molecule, essential for peptide antigen presentation to cytotoxic T cells.
- It is a secreted and cell surface protein, non-covalently associated with the MHC I heavy chain, and is present in body fluids and on nucleated cells.
- M. tuberculosis EsxA or EsxA-EsxB can bind B2M intracellularly, reducing its cell surface export and impairing class I antigen presentation, a mechanism of immune evasion (PubMed:25356553). Related references: PMID:25356553
- Structurally, it contains an immunoglobulin-like domain and features a disulfide bond, and it can undergo glycation and N-terminal pyroglutamate formation.
- Mutations in B2M are linked to hereditary amyloidosis, where amyloid fibrils deposit in tissues; the protein itself is prone to aggregation.
- The mature protein has a molecular weight of approximately 13.7 kDa.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 40–119 of human B2M. For Western blot, consider using reducing and non-reducing conditions to assess the disulfide-linked conformation.
- For immunofluorescence (IF-P) and immunohistochemistry (IHC-P), appropriate fixation and antigen retrieval may be required; validate in tissue sections known to express MHC I (e.g., lymph node, spleen).
- ELISA applications may use this antibody as a capture or detection reagent for B2M quantitation in serum or culture supernatants.
- Cross-reactivity with mouse and rat B2M suggests a conserved epitope; however, verify performance if studying non-human species.
