Biological Background: Beta-2-microglobulin Function and Localization
- Beta-2-microglobulin (B2M) is the light chain of the class I major histocompatibility complex (MHC), essential for presenting peptide antigens to the immune system. Exogenously applied M.tuberculosis EsxA decreases B2M export to the cell surface, potentially impairing class I antigen presentation. Related references: PMID:25356553
- It is secreted and displayed on the cell surface as a component of MHC I, bridging antigen-loaded heavy chains and CD8+ T cells.
- Structurally, B2M contains an immunoglobulin-like domain and a conserved disulfide bond; it is classified as a glycoprotein.
- The protein is linked to amyloidosis, forming amyloid fibrils in long-term hemodialysis patients; genetic variants are associated with disease susceptibility.
- Post-translational modifications include glycation and pyrrolidone carboxylic acid formation at the N-terminus.

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 21-119, the mature secreted form; verify antibody reactivity under both denaturing and native conditions for WB and FC.
- For WB, the predicted mass is ~13 kDa; use 15% acrylamide gels, optimize transfer time, and include low-molecular-weight markers.
- In IHC-P and IF/ICC, gentle fixation and antigen retrieval are recommended to preserve native cell surface localization.
- For FC, include proper isotype controls; B2M is expressed on most nucleated cells, so confirm specificity in target versus knockout/knockdown models.
- Cross-reactivity is indicated only for human; validate in other species if needed.
