Biological Background: BDNF Function and Localization
- BDNF (Brain-derived neurotrophic factor), also known as Abrineurin, is a secreted neurotrophic factor synthesized as a precursor protein.
- BDNF is a key signaling molecule that activates the NTRK2 receptor, promoting neuronal survival, differentiation, and axonal growth during development.
Related references:
PMID:11152678 - In the adult brain, BDNF is a major regulator of synaptic transmission and plasticity, including long-term potentiation (LTP) and long-term depression (LTD), and modulates dendritic growth and morphology.
- BDNF also signals through a heterodimeric receptor complex of NGFR and SORCS2, which mediates synaptic plasticity and can promote neuronal apoptosis.
Related references:
PMID:24908487, PMID:29909994 - BDNF is a secreted protein, acting in both autocrine and paracrine fashions.
- BDNF is detected in blood plasma and saliva at the protein level, and is highly expressed in the hippocampus, amygdala, cerebral cortex, and cerebellum, with additional expression in heart, lung, skeletal muscle, testis, prostate, and placenta.
Related references:
PMID:11152678, PMID:19467646 - BDNF is a glycoprotein with a signal peptide and disulfide bonds. It undergoes cleavage on pair of basic residues to generate the mature form. Associated keywords include Alternative promoter usage, Alternative splicing, and Proteomics identification.
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide corresponding to amino acids 100–200 of human BDNF. This region shows high conservation with mouse and rat BDNF, consistent with the observed cross-reactivity. For applications in other species, verify sequence homology before use.
- For immunofluorescence (IF-P), consider permeabilization methods appropriate for secreted proteins to retain proper localization.
- In ELISA applications, the antibody can be used as a capture or detection reagent. Pairing with a matched antibody pair is recommended for optimal assay performance.
- Due to BDNF's secretion, sample preparation from biological fluids (e.g., plasma, saliva) may require careful handling to preserve protein integrity.
