Biological Background: Bcl-XL Function and Localization

• Bcl-2-like protein 1 (Bcl-XL) is a key anti-apoptotic member of the Bcl-2 family, encoded by the BCL2L1 gene. Alternative splicing produces two major isoforms: Bcl-X(L), the long isoform that inhibits apoptosis, and Bcl-X(S), the short isoform that promotes apoptosis.
• Bcl-X(L) potently inhibits cell death by binding to the voltage-dependent anion channel (VDAC) on the mitochondrial outer membrane, preventing the release of cytochrome c and blocking caspase activation.
• Isoform Bcl-X(L) also plays a role in regulating presynaptic plasticity, including neurotransmitter release and recovery, mitochondrial ATP availability, and endocytic vesicle retrieval in hippocampal neurons through association with DNM1L and stimulation of its GTPase activity.
