Biological Background: Bcl-XL Function and Localization
- Bcl-2-like protein 1 (Bcl-XL) is a key anti-apoptotic member of the Bcl-2 family, encoded by the BCL2L1 gene. Alternative splicing produces two major isoforms: Bcl-X(L), the long isoform that inhibits apoptosis, and Bcl-X(S), the short isoform that promotes apoptosis.
- Bcl-X(L) potently inhibits cell death by binding to the voltage-dependent anion channel (VDAC) on the mitochondrial outer membrane, preventing the release of cytochrome c and blocking caspase activation.
- Isoform Bcl-X(L) also plays a role in regulating presynaptic plasticity, including neurotransmitter release and recovery, mitochondrial ATP availability, and endocytic vesicle retrieval in hippocampal neurons through association with DNM1L and stimulation of its GTPase activity. - Bcl-XL localizes to multiple subcellular compartments including the mitochondrial inner and outer membranes, mitochondrial matrix, cytoplasmic vesicles (e.g., synaptic vesicles), cytosol, centrosome, and nuclear membrane.
- The protein is expressed in a tissue-specific manner: Bcl-X(L) is abundant in long-lived postmitotic cells like adult brain, while Bcl-X(S) is found in cells with high turnover such as developing lymphocytes.
- Post-translational modifications and additional keywords include phosphorylation, ubiquitination, alternative splicing, and a transmembrane helix; these are critical for its regulation and function in apoptosis, endocytosis, and synaptic processes.
Experimental Guidance and Technical Tips
- The immunogen (aa1-120 of human Bcl-XL) spans a region common to both Bcl-X(L) and Bcl-X(S) isoforms; therefore, the antibody may recognize both forms. Consider validating isoform specificity using isoform-specific controls or detection methods.
- For Western blot, a predominant band near 26 kDa corresponding to Bcl-X(L) is expected, with possible additional bands for other isoforms or post-translational modifications. Optimize loading and transfer conditions, and use appropriate positive and negative cell lysates.
- This antibody is validated for Western blot and ELISA. For ELISA, consider using recombinant Bcl-XL protein or cell lysates, and include appropriate controls to ensure signal specificity.
- Given the involvement of Bcl-XL in apoptosis, expression levels may vary with cell treatment or disease state; thus, include treatment controls (e.g., apoptosis inducers) in experiments.
