Biological Background: Bcl-2 Function and Localization

  • Apoptosis regulator Bcl-2 suppresses apoptosis in a variety of cell systems, including factor-dependent lymphohematopoietic and neural cells.
  • It regulates cell death by controlling mitochondrial membrane permeability and functions in a feedback loop with caspases, inhibiting caspase activity by preventing cytochrome c release and/or binding APAF-1.
  • Bcl-2 also acts as an inhibitor of autophagy under non-starvation conditions by interacting with BECN1 and AMBRA1.
  • It may attenuate inflammation by impairing NLRP1-inflammasome activation, thereby reducing CASP1 activation and IL1B release.
  • Subcellularly, Bcl-2 localizes to the mitochondrial outer membrane, nucleus membrane, endoplasmic reticulum membrane, and cytoplasm.
  • Expressed in many tissues, with highest levels in reproductive tissues. In brain, it is found in specific neuronal populations like olfactory bulb mitral cells, hippocampal granule/pyramidal neurons, pontine nuclei, and cerebellar granule neurons, with higher prenatal expression in neuroepithelium and cortical plate.
  • Keywords: Apoptosis, Autophagy, Endoplasmic reticulum, Membrane, Mitochondrion, Mitochondrion outer membrane, Nucleus, Phosphoprotein, Reference proteome, Transmembrane, Transmembrane helix, Ubl conjugation.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide from the N-terminus (aa 1-100) of rat Bcl-2, which may contribute to cross-reactivity (confirmed for mouse and rat).
  • Suitable for Western blotting (WB), immunohistochemistry on paraffin sections (IHC-P), and ELISA. For IHC-P, consider antigen retrieval protocols appropriate for nuclear/membrane targets.
  • Always validate antibody performance in your specific sample system and include appropriate controls.
