Biological Background: Bcl-2 Function and Localization
- Apoptosis regulator Bcl-2 suppresses apoptosis in a variety of cell systems, including factor-dependent lymphohematopoietic and neural cells.
- It regulates cell death by controlling mitochondrial membrane permeability and functions in a feedback loop with caspases, inhibiting caspase activity by preventing cytochrome c release and/or binding APAF-1.
- Bcl-2 also acts as an inhibitor of autophagy under non-starvation conditions by interacting with BECN1 and AMBRA1.
- It may attenuate inflammation by impairing NLRP1-inflammasome activation, thereby reducing CASP1 activation and IL1B release.
- Subcellularly, Bcl-2 localizes to the mitochondrial outer membrane, nucleus membrane, endoplasmic reticulum membrane, and cytoplasm.
- Expressed in many tissues, with highest levels in reproductive tissues. In brain, it is found in specific neuronal populations like olfactory bulb mitral cells, hippocampal granule/pyramidal neurons, pontine nuclei, and cerebellar granule neurons, with higher prenatal expression in neuroepithelium and cortical plate.
