Biological Background: Bcl-2 Function and Localization
- Bcl-2, encoded by the Bcl2 gene, is a key anti-apoptotic protein that suppresses programmed cell death in various cell systems, including factor-dependent lymphohematopoietic and neural cells.
- It regulates cell death by controlling mitochondrial membrane permeability, functioning in a feedback loop with caspases. It inhibits caspase activity either by preventing cytochrome c release from mitochondria or by binding to the apoptosis-activating factor APAF-1.
- Beyond apoptosis, Bcl-2 acts as an inhibitor of autophagy: it interacts with BECN1 and AMBRA1 under non-starvation conditions to suppress autophagy function.
- It may also attenuate inflammation by impairing NLRP1-inflammasome activation, thereby reducing CASP1 activation and IL1B release.
- Subcellularly, Bcl-2 localizes to the mitochondrial outer membrane, nucleus membrane, endoplasmic reticulum membrane, and cytoplasm.
- Tissue expression is broad, with highest levels in reproductive tissues. In the adult brain, it is found in mitral cells of the olfactory bulb, hippocampal granule and pyramidal neurons, pontine nuclei, cerebellar granule neurons, and ependymal cells; prenatal brain expression is higher in the neuroepithelium and cortical plate.
- The protein has a calculated molecular weight of approximately 26.6 kDa and is subject to post-translational modifications such as phosphorylation and ubiquitination (Ubl conjugation).
Experimental Guidance and Technical Tips
- The immunogen includes a synthetic peptide corresponding to residues 1–100 of rat Bcl-2, suggesting the antibody recognizes the N-terminal region. This region may be conserved across species; cross-reactivity with mouse is confirmed, but testing in additional species should be validated.
- For Western blot (WB), load appropriate positive control lysates (e.g., from rat or mouse tissues/cells expressing Bcl-2) and expect a band at ~26 kDa. Reducing conditions are typically used, but optimization may be needed.
- For immunoprecipitation (IP), titrate the antibody amount and include relevant isotype controls. Pre-clearing lysates can reduce nonspecific binding.
- For ELISA, the antibody may be used as a capture or detection reagent; validate pairing with a suitable secondary antibody for your assay format.
- Always include appropriate negative controls to assess specificity, and consider testing in the endogenous system of interest.
