Biological Background: Bcl-2 Function and Localization
- Apoptosis regulator Bcl-2 (also known as PPP1R50) is encoded by the BCL2 gene and undergoes alternative splicing.
- Bcl-2 suppresses apoptosis in a variety of cell systems including factor-dependent lymphohematopoietic and neural cells. Related references: PMID:1508712 PMID:8183370
- It regulates cell death by controlling mitochondrial membrane permeability, functioning in a feedback loop with caspases, and inhibiting caspase activity by preventing cytochrome c release and binding APAF-1. Related references: PMID:11368354
- Bcl-2 also acts as an inhibitor of autophagy through interaction with BECN1 and AMBRA1 during non-starvation conditions. Related references: PMID:18570871 PMID:20889974 PMID:21358617
- It may attenuate inflammation by impairing NLRP1-inflammasome activation, thereby reducing CASP1 activation and IL1B release. Related references: PMID:17418785
- Bcl-2 localizes to the mitochondrial outer membrane, nuclear membrane, endoplasmic reticulum membrane, and cytoplasm.
- It is broadly expressed in a variety of tissues.
- Bcl-2 is a phosphoprotein subject to ubiquitin-like conjugation and is associated with chromosomal rearrangements in diseases such as lymphoma.
Experimental Guidance and Technical Tips
- This recombinant monoclonal antibody was generated using a synthetic peptide corresponding to human/rat Bcl-2; it may detect conserved epitopes across species.
- Validated applications include Western blot (WB), immunohistochemistry on paraffin-embedded sections (IHC-P), and ELISA.
- The antibody cross-reacts with human, mouse, and rat Bcl-2, making it suitable for multi-species studies.
- For optimal results, titrate the antibody and validate in your specific experimental system.
