Biological Background: BCKDK Function and Localization
- BCKDK (Branched-chain alpha-ketoacid dehydrogenase kinase) is a serine/threonine-protein kinase that forms a functional pair with the phosphatase PPM1K, serving as a metabolic regulatory node coordinating branched-chain amino acids (BCAAs) with glucose and lipid metabolism. Related references: PMID:24449431 PMID:29779826 PMID:37558654
- It phosphorylates the BCKDHA subunit of the mitochondrial branched-chain alpha-ketoacid dehydrogenase (BCKDH) complex on Ser-337, leading to inactivation of the complex and suppression of BCAA catabolism. Related references: PMID:29779826 PMID:37558654
- BCKDK also phosphorylates cytosolic ACLY (ATP citrate lyase) on Ser-455 in response to carbohydrate abundance, enhancing lipogenesis and glucogenesis during the fasting-to-feeding transition. Related references: PMID:29779826
- It recognizes phosphosite motifs with the canonical sequence SxxE/D. Related references: PMID:29779826
- Located in the mitochondrial matrix, BCKDK associates with the E2 component of the BCKDH complex.
- Expressed ubiquitously across tissues.
- UniProt keywords associate BCKDK with autism spectrum disorder, epilepsy, and intellectual disability, suggesting potential disease relevance.
- Post-translational modifications include acetylation and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1–100) of human BCKDK; for WB analysis, consider whether the epitope remains accessible in your specific sample preparation.
- Cross-reactivity with mouse and rat makes this antibody a candidate for translational metabolic studies in rodent models.
- For ELISA applications, validate the antibody in your target sample matrix and optimize blocking and washing conditions.
