Biological Background: BCAT2 Function and Localization
- BCAT2 (branched chain amino acid transaminase 2), also known as placental protein 18 (PP18), is a mitochondrial aminotransferase.
- Catalyzes the first reaction in the catabolism of the essential branched chain amino acids leucine, isoleucine, and valine. Related references: PMID:17050531, PMID:25653144, PMID:8702755
- Branched chain amino acid catabolism provides acetyl-CoA for lipid synthesis during adipocyte differentiation and may regulate adipose tissue browning by modulating PRDM16 acetylation.
- The enzyme is ubiquitously expressed and localizes to the mitochondrion.
- BCAT2 is a pyridoxal phosphate-dependent enzyme belonging to the class-IV aminotransferase family; post-translational modifications include acetylation.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 28 – 392 of human BCAT2, representing nearly the full-length protein, which supports high target specificity.
- In western blot, the predicted mitochondrial form migrates at ~44 kDa; additional bands (e.g., ~33 kDa) may reflect alternative isoforms or processing.
- The antibody has been reported for WB, IF-P, IHC-P, and ELISA; for each application, titrate and validate conditions in the relevant sample system.
- Cross-reactivity with mouse and rat extends utility to rodent models, but confirm band patterns in your model of interest.
