Biological Background: BCAS3 Function and Localization
- BCAS3 (Breast carcinoma-amplified sequence 3), also known as GAOB1, is a microtubule-associated cell migration factor that contains WD repeats and acts as a proto-oncogene.
- Plays a pivotal role in angiogenesis by regulating cell polarity and directional endothelial cell migration, mediating activation and recruitment of CDC42 and reorganization of the actin cytoskeleton at the leading edge.
- Functions synergistically with PELP1 as a transcriptional coactivator of estrogen receptor-responsive genes, stimulating histone acetyltransferase activity and binding to chromatin.
- Participates in autophagy regulation; in complex with PHAF1, it associates with preautophagosomal structures and likely binds phosphatidylinositol 3-phosphate (PtdIns3P) to mediate recruitment during both non-selective and selective autophagy. Related references: PMID:33499712
- Localizes to the nucleus, cytoplasm, cytoskeleton, and preautophagosomal structures, consistent with its roles in transcription, cytoskeletal dynamics, and autophagy.
- Broadly expressed in tissues including stomach, liver, lung, kidney, prostate, testis, thyroid, adrenal gland, brain, heart, skeletal muscle, colon, spleen, small intestine, placenta, blood leukocytes, and mammary epithelial cells; highly expressed in breast cancers and glioma cell lines.
