Biological Background: BCAR1 Function and Localization
- BCAR1 (Breast cancer anti-estrogen resistance protein 1, also known as p130cas, CAS, CASS1, CRKAS) is a docking protein and a member of the Cas scaffolding protein family. It exists in multiple isoforms due to alternative splicing.
- BCAR1 plays a central coordinating role in tyrosine kinase-based signaling pathways that regulate cell adhesion. It is implicated in the induction of cell migration and cell branching. Related references: PMID:12432078, PMID:12832404, PMID:17038317
- It is involved in the BCAR3-mediated inhibition of TGF-beta signaling (By similarity).
- Subcellularly, BCAR1 localizes to cell junctions, focal adhesions, the cytoplasm, and cell projections such as axons.
- As a phosphoprotein that undergoes acetylation, BCAR1 contains both an SH3 domain and SH3-binding motifs, facilitating protein-protein interactions essential for signal transduction.
- BCAR1 is widely expressed, with abundant expression in the testis and lower levels in the liver, thymus, and peripheral blood leukocytes. It is also expressed in B-cells at the protein level.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 495–612, a region within the central domain of BCAR1, which may detect multiple isoforms if the sequence is conserved.
- For Western blot, expect a band near 93 kDa; consider using cell lines with established BCAR1 expression (e.g., breast cancer lines) and include phosphatase inhibitors to preserve phosphorylation.
- In IHC-P, antigen retrieval conditions should be empirically optimized; use testis sections as a positive control due to high BCAR1 expression.
- For IF/ICC, fixation and permeabilization protocols need optimization to visualize BCAR1 at focal adhesions and in the cytoplasm.
- Cross-reactivity with mouse and rat enables translational studies; however, verify epitope conservation through sequence alignment.
