Biological Background: BAZ2A / TIP5 Function and Localization
- BAZ2A, also known as TIP5 or WALp3, is a large multidomain protein that serves as the regulatory subunit of the ATP-dependent NoRC-1 and NoRC-5 ISWI chromatin remodeling complexes.
- These complexes catalyze ordered nucleosome spacing and facilitate DNA access during replication, transcription, and repair, with BAZ2A directly stimulating the ATPase activity of SMARCA5 in the NoRC-5 complex (PubMed:28801535). Related references: PMID:28801535
- Within the nucleolus, BAZ2A mediates rDNA silencing by recruiting histone deacetylases (e.g., HDAC1) and DNA methyltransferases to establish repressive heterochromatin, thereby inhibiting RNA polymerase I transcription.
- Targeting to rDNA is achieved through interaction with the transcription termination factor TTF1 and specific recognition of histone H4 acetylated at lysine 16 (H4K16ac), which subsequently triggers deacetylation of adjacent H4 residues.
- BAZ2A also binds promoter-associated RNAs (pRNAs) complementary to the rDNA promoter, a step essential for heterochromatin formation and transcriptional silencing.
