Biological Background: BAZ2A Function and Localization
- BAZ2A, also known as TIP5 or hWALp3, is a regulatory subunit of the NoRC-1 and NoRC-5 ISWI chromatin remodeling complexes that form ordered nucleosome arrays and facilitate DNA access during replication, transcription, and repair. Related references: PMID:28801535
- It directly stimulates the ATPase activity of SMARCA5 and regulates nucleosome spacing, with ATP hydrolysis rates differing between NoRC-1 and NoRC-5 complexes. Related references: PMID:28801535
- In the NoRC-5 complex, BAZ2A mediates rDNA silencing by recruiting histone-modifying enzymes such as HDAC1 and DNA methyltransferases, leading to heterochromatin formation and transcriptional repression of RNA polymerase I.
- It is recruited to rDNA via interaction with TTF1 and recognition of acetylated histone H4K16ac, resulting in deacetylation of other H4 lysines (K5, K8, K12).
- BAZ2A specifically binds pRNAs (150-250 nt) complementary to the rDNA promoter, a step required for heterochromatin formation and gene silencing.
- Subcellular localization is predominantly nucleolar.
- Tissue expression is moderate and widespread, including heart, brain, placenta, lung, skeletal muscle, kidney, and pancreas.
- The protein features bromodomains, zinc-fingers, coiled-coil regions, and undergoes post-translational modifications such as acetylation, phosphorylation, and ubiquitylation.
