Biological Background: Bax Function and Localization
- Bax is a central apoptosis regulator that drives mitochondrial outer membrane permeabilization. Upon cellular stress, it undergoes a conformational change, translocates to the mitochondrial outer membrane, and triggers cytochrome c release, which activates caspase-3 and downstream apoptosis. - Under normal conditions, Bax is maintained in an inactive state largely in the cytosol through constant retrotranslocation from mitochondria to the cytosol mediated by BCL2L1/Bcl-xL, preventing toxic mitochondrial accumulation. Related references: PMID:21458670
- Bax is expressed in a wide variety of tissues. Multiple isoforms are generated by alternative splicing: isoform Psi is found in glial tumors; isoform Alpha is expressed in spleen, breast, ovary, testis, colon and brain, with low levels in skin and lung; isoform Sigma shows a similar tissue distribution and is also present in many cancer cell lines.
- Subcellularly, Bax localizes to the mitochondrion outer membrane, cytoplasm, and nucleus, with its distribution shifting in response to apoptotic signals.
- Post-translational modifications such as acetylation, Ubl conjugation, and isopeptide bond formation may modulate Bax activity and stability.

Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–100) of human Bax, which is highly conserved across human, mouse, and rat, likely contributing to the observed cross-reactivity. When validating in your model system, confirm specificity via knockdown or knockout controls.
- For western blot (WB), expect a band at ~21 kDa. Use reducing conditions and appropriate loading controls; consider that multiple isoforms may produce additional bands.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval and fixation methods can influence detection of cytosolic versus mitochondrial Bax pools. Pilot experiments are advised to optimize conditions.
- This polyclonal antibody may be suitable for immunoprecipitation (IP) and ELISA. For ELISA, the peptide immunogen can serve as a standard for quantitation.
