Biological Background: Bax Function and Localization
- BAX is a pro-apoptotic member of the Bcl-2 protein family, critical for the intrinsic apoptosis pathway.
- Under normal conditions, BAX is largely cytosolic, with constant retrotranslocation from mitochondria mediated by BCL2L1/Bcl-xL, preventing toxic accumulation at the mitochondrial outer membrane (MOM). Related references: PMID:21458670
- Upon stress signals, BAX undergoes a conformational change, translocates to the MOM, and facilitates cytochrome c release, leading to CASP3 activation and apoptosis. Related references: PMID:10772918 PMID:11060313 PMID:16113678
- Localizes to the cytoplasm, mitochondrial outer membrane, and nucleus, with its subcellular distribution tightly regulated.
- Multiple isoforms exist (Alpha, Sigma, Psi) with tissue-specific expression patterns; isoform Psi is enriched in glial tumors.
- Post-translational modifications include acetylation and ubiquitination (isopeptide bond formation), modulating its activity and stability.
- BAX is widely expressed across tissues and functions as a tumor suppressor; its dysregulation is implicated in various cancers.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-100) of human Bax; align sequences with mouse and rat to confirm cross-reactivity.
- For Western blot (WB), a band near 21 kDa is expected; post-translational modifications may cause minor shifts.
- For IF/ICC, validate mitochondrial and cytoplasmic staining; include controls for apoptosis-induced translocation assays.
- For ELISA, optimize coating and detection conditions; a paired antibody approach may be suitable for sandwich ELISA development.
