Biological Background: BAT3/BAG6 Function and Localization
- Protein identity and gene family: Large proline-rich protein BAG6, also known as BAG family molecular chaperone regulator 6 (BAG-6), HLA-B-associated transcript 3 (BAT3), Protein G3, and Scythe. Encoded by the BAG6 gene (GeneID: 7917) and belonging to the BAG family of co-chaperones.
- Chaperone-mediated protein quality control: Functions as an ATP-independent molecular chaperone that prevents aggregation of misfolded and hydrophobic proteins, acting as part of the BAG6/BAT3 complex to maintain client proteins soluble for delivery to the endoplasmic reticulum or targeting to the proteasome. Related references: PMID:21636303, PMID:20516149, PMID:21743475
- Tail-anchored protein delivery: Recruited to ribosomes, interacts with the transmembrane region of newly synthesized tail-anchored proteins, and together with SGTA and ASNA1 mediates their delivery to the ER. Related references: PMID:20516149, PMID:20676083, PMID:25535373
- Ubiquitin-dependent sorting and degradation: Client proteins that fail ER delivery are ubiquitinated by RNF126 and sorted to the proteasome; SGTA negatively regulates this process by preventing RNF126 recruitment. Related references: PMID:24981174, PMID:27193484, PMID:28104892, PMID:23129660, PMID:25179605
- Endoplasmic reticulum stress response: Involved in pre-emptive quality control during ER stress, rerouting nascent proteins to the cytosol for proteasomal degradation to protect the ER from overload. Related references: PMID:26565908
- DNA damage and chromatin regulation: In response to DNA damage, accumulates in the nucleus, forms a complex with p300/EP300 to enhance p53 acetylation and transcriptional activity. May also regulate H3K4 dimethylation as part of a chromatin regulator complex. Related references: PMID:17403783, PMID:18765639
- Immune function and secretion: Released extracellularly via exosomes, acts as a ligand for NK cell receptor NCR3, stimulating NK cell cytotoxicity against tumor cells and immature dendritic cells.
- Subcellular localization and tissue specificity: Distributed in the cytosol, nucleus, and secreted via exosomes. Expressed at protein level in immature dendritic cells. Key post‑translational modifications include acetylation, phosphorylation, and ubiquitination.
Experimental Guidance and Technical Tips
- Immunogen Design: The immunogen corresponds to the N‑terminal region (amino acids 1‑200) of human BAT3/BAG6. This region may be conserved across species, consistent with observed cross‑reactivity with mouse and rat.
- Western Blotting (WB): Consider using whole‑cell lysates from human, mouse, or rat cell lines as positive controls. The predicted molecular weight is ~119 kDa; validate band specificity with appropriate blocking and detection conditions.
- ELISA Applications: For sandwich ELISA development, consider pairing with a matched monoclonal antibody targeting a distinct epitope. Validate assay performance with recombinant protein standards.
- General Note: Always validate antibody performance in your specific experimental system, as optimal dilutions and conditions may vary.
