Biological Background: BAP31 Function and Localization
- BAP31 (BCAP31) is a 28 kDa endoplasmic reticulum (ER) membrane protein, also known as 6C6-AG tumor-associated antigen and protein CDM/p28.
- Functions as a chaperone and is one of the most abundant ER proteins, playing a key role in the export of secreted proteins and the recognition/targeting of misfolded proteins for ER-associated degradation (ERAD). Related references: PMID:18287538 PMID:9396746
- Serves as a cargo receptor for the export of transmembrane proteins (By similarity).
- Plays a role in mitochondrial Complex I assembly by stimulating the translocation of NDUFS4 and NDUFB11 from the cytosol to the mitochondria via interaction with TOMM40. Related references: PMID:31206022
- Upon ER stress, BAP31 delocalizes from ER-mitochondria contact sites and binds BCL2; may be involved in CASP8-mediated apoptosis. Related references: PMID:31206022 PMID:10958671
- Subcellularly, BAP31 localizes to the endoplasmic reticulum membrane and the ER-Golgi intermediate compartment membrane.
- Ubiquitously expressed, with high expression in neurons and discrete endocrine cells.
- Genetic variants in BCAP31 are linked to disorders such as deafness, dystonia, and intellectual disability.
- Alternative splicing generates multiple isoforms, and the protein features a coiled-coil domain and transmembrane helices.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 127–246 of human BAP31; confirm epitope accessibility in your assay format.
- For western blot, a specific band near 28 kDa is expected; validate detection conditions using positive control lysates from human, mouse, or rat cells.
- For immunofluorescence/ICC, consider fixation and permeabilization protocols suitable for ER membrane proteins.
- The antibody is suitable for immunoprecipitation and ELISA applications; optimize concentrations and buffer conditions for each assay.
- Cross-reactivity with mouse and rat BAP31 is reported; verify performance in your specific model system.
