Biological Background: BAP31 Function and Localization
- BCAP31, also known as BAP31, CDM, 6C6-AG tumor-associated antigen, and p28, is a 28 kDa transmembrane protein.
- It is one of the most abundant proteins of the endoplasmic reticulum (ER) and functions as a molecular chaperone. Related references: PMID:18287538 PMID:9396746
- BAP31 plays a critical role in ER protein quality control by recognizing misfolded proteins and targeting them to ER-associated degradation (ERAD). Related references: PMID:18287538 PMID:9396746
- It also participates in the export of secretory proteins and acts as a cargo receptor for transmembrane proteins at the ER-Golgi intermediate compartment.
- Beyond its ER roles, BAP31 contributes to mitochondrial homeostasis by facilitating the assembly of respiratory chain Complex I. It interacts with TOMM40 to mediate the mitochondrial translocation of NDUFS4 and NDUFB11. Related references: PMID:31206022
- Under ER stress, BAP31 redistributes from ER-mitochondria contact sites and associates with BCL2, linking ER stress to apoptotic signaling. Related references: PMID:31206022
- BAP31 may also be involved in CASP8-mediated apoptosis, further underscoring its role in cell death pathways. Related references: PMID:10958671
- BAP31 is ubiquitously expressed, with particularly high levels in neurons and endocrine cells. Mutations in BCAP31 are linked to X-linked disorders including deafness, dystonia, and intellectual disability.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering the central region (aa 101-200) of human BAP31, which may contribute to the antibody's cross-reactivity with mouse and rat orthologs.
- For Western blot (WB), the predicted molecular weight is 28 kDa; however, post-translational modifications or dimerization may result in additional bands—validate in the relevant sample system.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval and permeabilization conditions may need optimization due to the transmembrane nature of BAP31.
- ELISA applications should consider coating with recombinant BAP31 protein or cell lysates; proper negative controls are recommended to ensure specificity.
