Biological Background: BAIAP2 Function and Localization
- BAIAP2 (BAI1-associated protein 2), also known as IRSp53, FLAF3, BAP2, and WAML, is a BAR/IMD domain-containing adapter protein. It exists in multiple isoforms due to alternative splicing; isoform 1 and isoform 4 are predominantly expressed in the brain, while a short isoform is ubiquitously expressed with highest levels in liver, prostate, testis, and placenta.
- Core Function: BAIAP2 links membrane-bound small G proteins (CDC42, RAC1) to cytoplasmic effector proteins, regulating actin cytoskeleton reorganization. It is required for CDC42-mediated actin reorganization and RAC1-driven membrane ruffling, and works with WASF family proteins and the Arp2/3 complex to remodel actin.

- The immunogen corresponds to a C-terminal region (aa 362-521) of human BAIAP2; this region may be conserved across species, enabling detection of human, mouse, and rat orthologs.
- For Western blotting, the expected molecular weight is approximately 61 kDa; however, multiple bands corresponding to splice isoforms or phospho-forms may appear. Titrate antibody concentration and validate in relevant tissue or cell lysates (e.g., brain extracts).
- In immunocytochemistry/immunofluorescence (IF/ICC), expect staining at filopodia, membrane ruffles, and cytoplasm. Fixation and permeabilization conditions should be optimized for your cell type.
- For ELISA, the antibody can be used as a capture or detection reagent; pair with a matched antibody or use individually for antigen detection, and validate with recombinant BAIAP2 protein or peptide.
