Biological Background: BAG5 Function and Localization

  • BAG5, also known as Bcl-2-associated athanogene 5 or BAG-5, is a member of the BAG domain-containing protein family that functions as co-chaperones.
  • It acts as a nucleotide-exchange factor for HSP70/HSPA proteins, accelerating ADP release and driving the chaperone cycle to promote protein refolding.

  • At junctional membrane complexes (JMC), BAG5 is essential for proteostasis, likely by scaffolding between HSPA8 chaperone and JMC proteins to facilitate proper folding.
  • BAG5 inhibits the E3 ubiquitin-protein ligase PRKN, suppressing both auto-ubiquitination of PRKN and ubiquitination of its substrate proteins.
  • Primarily expressed in the heart, BAG5 is linked to cardiomyopathy; disease-associated variants have been identified.
  • The protein undergoes alternative splicing and contains multiple BAG domains.
  • The full-length protein has a molecular weight of approximately 51-52 kDa.

Experimental Guidance and Technical Tips

  • The immunogen (amino acids 198-447) targets the C-terminal portion of BAG5, including functional BAG domains; this design may detect full-length and certain splice variants.
  • For Western blotting, the predicted band is ~51-52 kDa; verify in relevant tissue/cell lysates (e.g., heart samples) and consider using a positive control expressing BAG5.
  • In immunohistochemistry (IHC-P), optimize antigen retrieval and test appropriate antibody dilutions; cardiac muscle sections may serve as a positive control given heart expression.
  • ELISA applications can use recombinant BAG5 protein or peptide fragments for standard curve generation and antibody titer determination.
  • As a polyclonal antibody, lot-to-lot consistency should be evaluated; include appropriate negative controls to confirm specificity.
