Biological Background: BACH1 Function and Localization

  • Transcription regulator BACH1 (BTB and CNC homolog 1, also known as HA2303) is a member of the CNC-bZIP transcription factor family.
  • Contains a BTB domain and a basic leucine zipper (bZIP) domain, enabling DNA binding and protein-protein interactions.
  • Acts as a context-dependent repressor or activator, binding NF-E2 DNA binding sites and coordinating transcription with MAFK (By similarity).
  • Together with MAF, represses transcription of genes under the control of the NFE2L2 oxidative stress pathway, thereby regulating cellular redox balance. Related references: PMID:24035498
  • Localizes predominantly to the nucleus, consistent with its function as a transcription regulator.
  • Subject to post-translational modifications such as phosphorylation and ubiquitination, which may influence its activity and stability.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1-320) of human BACH1, which includes the BTB domain, critical for protein interactions; this region is highly conserved in mouse, supporting cross-reactivity.
  • For Western Blot, expect a band at approximately 82 kDa; use whole-cell lysates from human or mouse cell lines and validate with positive controls expressing BACH1.
  • In Immunoprecipitation, the polyclonal nature may enhance capture efficiency; optimize antibody and lysate concentrations, and confirm with appropriate controls.
  • For ELISA, the antibody may be used for capture or detection; validate with recombinant BACH1 protein or relevant biological samples.
  • Consider performing a titration series to determine optimal working concentrations for your specific experimental setup.
