Biological Background: BACE1 Function and Localization
- BACE1 (Beta-secretase 1), also known as ASP2 or memapsin-2, is a type I transmembrane aspartyl protease belonging to the peptidase A1 family, encoded by the BACE1 gene on chromosome 11.
- BACE1 is responsible for the proteolytic processing of the amyloid precursor protein (APP), cleaving at the N-terminus of the Aβ peptide sequence to generate beta-cleaved soluble APP and a C-terminal fragment that is subsequently processed by gamma-secretase to produce amyloid-beta peptides, which are central to Alzheimer's disease pathology.

- BACE1 localizes to multiple cellular membranes, including the cell surface, Golgi apparatus, trans-Golgi network, endoplasmic reticulum, endosomes (early, late, recycling), lysosomes, and membrane rafts, and is enriched in neuronal projections like axons and dendrites.

- The immunogen for this antibody is a synthetic peptide corresponding to the cytoplasmic domain (amino acids 402-501) of human BACE1, a region unique to BACE1 and not conserved with BACE2. This design may confer specificity to the BACE1 isoform.
- For Western blot (WB), expect detection of full-length BACE1 at approximately 56 kDa. Use appropriate positive controls such as brain tissue lysates or cell lines known to express BACE1. Consider validating specificity with knockout or knockdown samples.
