- B9 domain-containing protein 1 (B9D1), also known as MKS1-related protein 1, is encoded by the B9D1 gene (GeneID: 27077).
- B9D1 is a component of the tectonic-like complex, which localizes at the transition zone of primary cilia and acts as a barrier to prevent diffusion of transmembrane proteins between the cilia and plasma membranes.
- It is required for ciliogenesis and sonic hedgehog (SHH) signaling.
- Subcellularly, B9D1 localizes to the cytoplasm, specifically at the cilium basal body and cilium axoneme.
- Mutations in B9D1 are associated with ciliopathies, including Joubert syndrome and Meckel syndrome.
