Biological Background: B4GALT4 Function and Localization
- B4GALT4 (Beta-1,4-galactosyltransferase 4) is a 40 kDa glycosyltransferase that catalyzes the transfer of galactose from UDP-Gal to N-acetylglucosamine via a β1-4 linkage, generating terminal N-acetyllactosamine units on glycoproteins and glycosphingolipids. Related references: PMID:12511560 PMID:17690104 PMID:32827291
- In keratan sulfate biosynthesis, B4GALT4 cooperates with B3GNT7 and sulfotransferases to construct and elongate mono- and disulfated disaccharide units. Related references: PMID:17690104
- It is involved in the synthesis of 6-sulfo-sialyl Lewis X (sLex) epitopes on mucin-type O-glycans, which serve as ligands for L-selectin and regulate leukocyte migration. Related references: PMID:12511560
- In neolacto-series glycosphingolipid biosynthesis, B4GALT4 transfers galactose to lactotriaosylceramide to form neolactotetraosylceramide. Related references: PMID:9792633
- B4GALT4 is localized to the Golgi apparatus membrane and can be secreted.
- It is expressed at highest levels in placenta, pancreas, kidney, and heart, and is also found in corneal epithelial cells. Related references: PMID:9792633 PMID:17690104
- The enzyme requires manganese as a cofactor and contains disulfide bonds and glycosylation sites, indicative of its post-translational modifications.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 35-344, covering a large portion of the protein, which may enhance antibody reactivity across multiple epitopes.
- For Western blotting, a band around 40 kDa is expected; consider using appropriate loading controls and post-translational modification-aware protocols.
- For ELISA, the antibody may be used for detection or capture; validate with relevant recombinant protein or cell lysates expressing B4GALT4.
- Since B4GALT4 is a Golgi-resident enzyme, subcellular fractionation or enrichment may improve signal detection in cell lines with low endogenous expression.
