Biological Background: B4GALT1 Function and Localization

  • B4GALT1 encodes beta-1,4-galactosyltransferase 1, a key glycosyltransferase that catalyzes the transfer of galactose from UDP-galactose to terminal N-acetylglucosamine residues on glycoproteins and glycolipids in the Golgi apparatus, essential for complex N-glycan biosynthesis and antibody Fc glycosylation. Related references: PMID:16157350, PMID:27872474, PMID:29133956, PMID:36280670
  • In conjunction with alpha-lactalbumin (LALBA), B4GALT1 forms the lactose synthase complex in the mammary gland, switching its acceptor specificity from GlcNAc to glucose to produce lactose during lactation. Related references: PMID:16157350
  • The protein exists in multiple forms: a Golgi-resident transmembrane form, a cell surface form involved in cell–cell and cell–matrix adhesion, and a secreted form capable of extracellular glycan remodeling.
  • B4GALT1 is ubiquitously expressed across tissues but shows very low levels in fetal and adult brain, highlighting tissue-specific glycosylation patterns.
  • Clinically, mutations in B4GALT1 are associated with congenital disorder of glycosylation type IId (CDG2D), emphasizing its role in human health and disease.

Experimental Guidance and Technical Tips

  • The immunogen targets the N-terminal catalytic domain (aa 45-210), which is conserved across known isoforms. Consider using this region for immunoassay design to ensure broad isoform recognition.
  • For Western blotting, expect a primary band near 44 kDa. Glycosylated or processed variants may appear at slightly higher molecular weights; validate in relevant human cell lysates.
  • This antibody is KO validated, indicating specific detection of B4GALT1. For critical applications, confirm specificity using B4GALT1 knockout cell lines or tissues.
  • ELISA applications may require antibody titration; test optimal coating and detection conditions in the intended sample matrix.
