Biological Background: B-Raf Function and Localization
- B-Raf is a serine/threonine-protein kinase that functions as a key transducer of the RAS-RAF-MEK-ERK mitogenic signaling pathway by phosphorylating MAP2K1/MEK1 (PubMed:21441910, PubMed:29433126).
Related references:
PMID:21441910, PMID:29433126 - It also phosphorylates PFKFB2, thereby integrating growth factor signaling with cellular glucose metabolism (PubMed:36402789).
Related references:
PMID:36402789 - B-Raf may contribute to synaptic plasticity, as suggested by its role in postsynaptic responses of hippocampal neurons (PubMed:1508179).
Related references:
PMID:1508179 - The V600E mutation (a valine to glutamate substitution at position 600) within the kinase domain leads to constitutive kinase activity and is a common driver in melanoma, colorectal, and thyroid cancers.
- B-Raf localizes to the nucleus, cytoplasm, and cell membrane, consistent with its role in transmitting extracellular signals to intracellular effectors.
- The protein is predominantly expressed in brain and testis and is subject to regulatory modifications including acetylation, methylation, and ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide covering amino acids 507–606, which includes the V600E mutation site; this design may confer specificity for the mutant over wild-type B-Raf.
- For Western blotting, use a standard SDS-PAGE gel optimized for an ~84 kDa protein and validate detection using lysates from cells expressing either wild-type or V600E B-Raf.
- In ELISA, consider including the immunizing peptide as a positive control to confirm binding specificity.
- As a polyclonal antibody, lot-to-lot variability is possible; we recommend confirming each new lot’s performance in your specific assay system.
