Biological Background: B-Raf Function and Localization

  • B-Raf is a serine/threonine-protein kinase encoded by the BRAF gene (also known as BRAF1, RAFB1), and is a member of the RAF family of proto-oncogenes. It is also referred to as proto-oncogene B-Raf, p94, or v-Raf murine sarcoma viral oncogene homolog B1.
  • A key component of the RAS-RAF-MEK-ERK signal transduction cascade, B-Raf transmits mitogenic signals from the cell membrane to the nucleus by phosphorylating and activating MAP2K1 (MEK1), thereby promoting cell proliferation and survival. Related references: PMID:21441910, PMID:29433126
  • B-Raf also phosphorylates additional substrates, such as PFKFB2, linking it to metabolic regulation. Related references: PMID:36402789
  • B-Raf is expressed predominantly in brain and testis, and may function in postsynaptic responses in hippocampal neurons. Related references: PMID:1508179
  • Subcellularly, B-Raf localizes to the nucleus, cytoplasm, and cell membrane, consistent with its role in signal transduction.
  • B-Raf undergoes multiple post-translational modifications, including phosphorylation, acetylation, and methylation, and contains a zinc-finger domain critical for its regulation and oncogenic activity.
  • Mutations in BRAF are associated with various diseases, such as cardiomyopathies, deafness, ectodermal dysplasia, and intellectual disability, highlighting its broad physiological importance.

Experimental Guidance and Technical Tips

  • As a polyclonal antibody raised against a recombinant fragment (aa 153-313), this product may recognize multiple epitopes and splice variants of B-Raf, offering high sensitivity.
  • Suitable applications include Western blotting (WB) and ELISA under standard conditions; optimal working dilutions should be determined empirically.
  • For WB, the expected molecular weight is approximately 84 kDa; consider using brain or testis tissue lysates as positive controls due to high B-Raf expression.
  • When using mouse or rat samples, verify cross-reactivity with a specific validation experiment, as cross-reactivity is listed but not guaranteed for all applications.
  • Polyclonal antibodies may exhibit lot-to-lot variability; we recommend routine validation and consistency testing for critical assays.
