Biological Background: AZI2 Function and Localization

  • AZI2 (5-azacytidine-induced protein 2), also known as NAP1 (Nak-associated protein 1) or TILP, is a 44935 Da adapter protein encoded by the AZI2 gene.
  • Functions as an adapter that binds the serine/threonine-protein kinase TBK1 and IKBKE, activating TBK1 and facilitating its oligomerization. Related references: PMID:14560022 PMID:21931631
  • Enhances TBK1-mediated phosphorylation of NF-kappa-B p65 subunit RELA and promotes NF-kappa-B activation induced by TBK1, TNF, or PMA. Related references: PMID:14560022 PMID:21931631
  • Participates in antiviral innate immunity by contributing to IFNB promoter activation via TICAM1. Related references: PMID:15611223
  • Localizes predominantly to the cytoplasm.
  • Widely expressed across tissues, with abundant levels observed in the pancreas and testis. Related references: PMID:14560022
  • Undergoes alternative splicing and is subject to phosphorylation and ubiquitin-like conjugation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a C-terminal region (aa 282-387) of human AZI2; consider this when designing detection antibodies for sandwich ELISA.
  • For Western blotting, a band at approximately 45 kDa is expected; validate in your sample system, noting that AZI2 is widely expressed.
  • Given cross-reactivity with mouse and rat, this antibody may be suitable for studies in these species; however, confirm reactivity with your specific model.
  • ELISA applications can be explored; optimal antibody concentration and buffer conditions should be determined experimentally.
