Biological Background: Axin-1 Function and Localization
- Axin-1 (Axis inhibition protein 1, also known as hAxin) is a scaffold protein encoded by the AXIN1 gene and belongs to the axin family; it acts as a tumor suppressor and developmental regulator.
- Functions as a core component of the beta-catenin destruction complex, where it facilitates phosphorylation and subsequent ubiquitination of CTNNB1, negatively modulating the canonical Wnt signaling pathway. Related references: PMID:12192039, PMID:27098453, PMID:28829046
- Controls dorsoventral patterning by both inhibiting CTNNB1/Wnt signaling and activating a Wnt-independent JNK pathway. Related references: PMID:12192039
- Enhances TGF-beta signaling by recruiting the RNF111 ubiquitin ligase to promote degradation of inhibitory SMAD7. Related references: PMID:16601693
- Forms a complex with HIPK2 and TP53, contributing to cell growth, apoptosis, and development; facilitates TP53 phosphorylation upon ultraviolet irradiation. Related references: PMID:17210684
- Serves as an adapter for AMPK activation in response to calorie restriction by mediating the association between STK11 and AMPK subunits at the late endosome membrane.
- Subcellularly localizes to the cytoplasm, nucleus, membrane, and late endosome; ubiquitously expressed in human tissues.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-351, covering the N-terminal region of human AXIN1; it may recognize full-length AXIN1 and potential isoforms.
- For Western blotting, a band around 96 kDa is expected; validate the detected band size and specificity in your relevant cell or tissue lysates.
- Optimize antibody dilution and blocking conditions for your individual assay setup; consider using AXIN1 knockout or overexpression controls to confirm specificity.
- In ELISA applications, confirm linearity of detection across a range of antigen concentrations and assess cross-reactivity with closely related proteins if needed.
