Biological Background: Advillin Function and Localization
- Advillin (AVIL), also known as p92, DOC6, ADVIL, and NPHS21, is a member of the gelsolin/villin family of actin regulatory proteins.
- It is a Ca²⁺-regulated actin-binding protein that plays a critical role in actin bundling and is required for various cellular processes, including ciliogenesis and the formation of actin-rich filopodia-like structures in fibroblasts. Related references: PMID:29058690
- Advillin is essential for neuronal morphogenesis; it mediates SREC1-regulated neurite-like outgrowth and contributes to regenerative sensory axon outgrowth and remodeling following peripheral injury in neonates. Related references: PMID:29058690
- In podocytes, advillin controls lamellipodia formation by modulating EGF-induced diacylglycerol generation through PLCE1 and ARP2/3 complex assembly, highlighting its role in kidney filtration barrier maintenance. Related references: PMID:29058690
- Subcellularly, advillin localizes to the cytoplasm and cytoskeleton, and is enriched at cell projections such as lamellipodia, neuron projections, axons, and focal adhesions.
- Tissue expression is highest in the small intestine and colonic lining, with weaker expression in thymus, prostate, testes, and uterus; it is also expressed in podocytes.
- Post-translational modifications include phosphorylation, and alternative splicing generates multiple isoforms, with the UniProt-reviewed 92 kDa canonical form serving as the immunogen for this antibody.
Experimental Guidance and Technical Tips
- The immunogen spans advillin residues 470–819, a region that is partially conserved across human, mouse, and rat, supporting the observed cross-reactivity.
- For Western blot (WB), a band of approximately 92 kDa is expected; however, due to alternative splicing and potential phosphorylation, additional bands may be observed. Validate with appropriate positive controls (e.g., lysates from small intestine or neuronal cells).
- In immunofluorescence/immunocytochemistry (IF/ICC), advillin staining is anticipated at lamellipodia, focal adhesions, and neuronal projections; optimization of fixation and permeabilization for cytoskeletal targets is advisable.
- For ELISA applications, consider using recombinant advillin as a standard and pair with a matched detection antibody for sandwich assays.
- Since the antibody is a polyclonal preparation, it may detect multiple advillin isoforms; lot-to-lot consistency should be verified in the intended application.
