Biological Background: AUTS2 Function and Localization
- AUTS2 (Autism susceptibility gene 2 protein, also known as MRD26 or FBRSL2) is a component of a Polycomb group (PcG) multiprotein PRC1-like complex that maintains the transcriptionally repressive state of many developmental genes, including Hox genes, through chromatin remodeling and histone H2A Lys-119 monoubiquitination. - In the PRC1-like complex containing PCGF5, RNF2, CSNK2B, RYBP and AUTS2, the ubiquitination activity is decreased due to RNF2 phosphorylation by CSNK2B, leading to transcriptional activation.
- In the cytoplasm, AUTS2 promotes axon and dendrite elongation and neuronal migration during embryonic brain development by reorganizing the actin cytoskeleton and activating RAC1 via interaction with RAC guanine nucleotide exchange factors (By similarity).
- AUTS2 localizes to the nucleus, cytoplasm, cytoskeleton, and cell projections including growth cones.
- It is strongly expressed in brain, skeletal muscle, and kidney, with additional expression in placenta, lung, and leukocytes.
- The protein is subject to alternative splicing and phosphorylation, and is associated with chromosomal rearrangements in autism spectrum disorder and intellectual disability.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 550-770 of human AUTS2, a central region not overlapping with known functional domains; consider using this antibody to detect full-length and potentially truncated isoforms.
- For Western blot, validate the antibody using positive control lysates from brain or skeletal muscle tissues, as AUTS2 is strongly expressed there; the expected band is ~139 kDa, but alternative splicing may produce additional bands.
- For immunocytochemistry/immunofluorescence, permeabilization is required to detect both nuclear and cytoplasmic pools; growth cone localization may be observed in neuronal models.
- In ELISA applications, consider using the recombinant protein immunogen as a positive control for sensitivity testing.
- Although the antibody cross-reacts with mouse and rat, confirm sequence homology in the immunogen region for your specific experimental model.
