Biological Background: ATXN2 Function and Localization
- ATXN2 encodes ataxin-2, also known as spinocerebellar ataxia type 2 protein and trinucleotide repeat-containing gene 13 protein.
- The primary function of ATXN2 involves negative regulation of endocytic EGFR internalization at the plasma membrane, influencing receptor trafficking.
- Ataxin-2 localizes to the cytoplasm, consistent with its role in intracellular trafficking and signaling.
- Tissue expression is broad, with highest levels in brain. Isoform 1 predominates in the brain and spinal cord, while isoform 4 is more abundant in the cerebellum. Within the brain, it is detected in the amygdala, caudate nucleus, corpus callosum, hippocampus, hypothalamus, substantia nigra, subthalamic nucleus, and thalamus.
- ATXN2 is subject to phosphorylation, ubiquitination, and methylation, and its dysfunction via CAG trinucleotide repeat expansion is linked to spinocerebellar ataxia type 2, amyotrophic lateral sclerosis, and Parkinsonism.
- Alternative splicing generates multiple isoforms, which may exhibit tissue-specific expression patterns.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 1250–1331 of human ATXN2. Verify target detection across relevant isoforms in your sample system.
- For Western blotting, ATXN2 has a predicted molecular weight of ~140 kDa; consider using a 6–8% SDS-PAGE gel and optimized transfer conditions for high-molecular-weight proteins.
- In immunohistochemistry (IHC-P), validate the antibody in brain tissue sections where ATXN2 is known to be highly expressed. Include appropriate negative controls.
- For immunofluorescence/ICC, expect a cytoplasmic staining pattern. Co-staining with organelle markers may help confirm localization.
