Biological Background: ATXN10 Function and Localization

  • Ataxin-10 (ATXN10) is also known as brain protein E46 homolog and spinocerebellar ataxia type 10 protein.
  • ATXN10 may play a role in the regulation of cytokinesis, as indicated by functional studies. Related references: PMID:21857149 PMID:25666058
  • It may also play a role in signaling by stimulating protein glycosylation and induces neuritogenesis by activating the Ras-MAP kinase pathway, which is necessary for the survival of cerebellar neurons (By similarity).
  • ATXN10 is expressed in the central nervous system.
  • Subcellular localization includes the cytoplasm, perinuclear region, midbody, centrosome, centriole, and cilium basal body.
  • Key post-translational modifications include phosphorylation, methylation, and Ubl conjugation. It is also subject to alternative splicing.
  • Mutations in ATXN10 are associated with spinocerebellar ataxia type 10, a neurodegenerative disorder.

Experimental Guidance and Technical Tips

  • For Western blot, a band at approximately 53 kDa is expected; consider using a neuronal cell line like SH-SY5Y as a positive control.
  • In immunofluorescence/ICC, expect cytoplasmic staining with possible enrichment at the perinuclear region, midbody, and centrosome; permeabilization is required.
  • When performing ELISA, titrate the antibody to determine the optimal working concentration for your specific assay format.
  • Validate the antibody in your relevant sample system to confirm reactivity, as cross-reactivity with mouse and rat has been reported.
