Biological Background: ATRX Function and Localization

  • ATRX (Transcriptional regulator ATRX, X-linked helicase II, XNP, Znf-HX) is a chromatin remodeler encoded by the ATRX gene (RAD54L, XH2) in humans.
  • Plays a central role in transcriptional regulation and chromatin remodeling; facilitates DNA replication at specific genomic loci and binds DNA tandem repeats and G4 quadruplex structures, incorporating H3.3-containing nucleosomes to stabilize these regions.
  • Functions as the catalytic subunit of the ATRX:DAXX complex, which exhibits ATP-dependent DNA translocase activity and mediates replication-independent deposition of histone H3.3 at pericentric repeats and telomeres.
  • Conflicting evidence exists regarding its role in TERRA regulation: one study indicates it is not sufficient to increase TERRA expression or decrease telomeric chromatin condensation in fibroblasts. Related references: PMID:24500201
  • ATRX forms a complex with ZNF274, TRIM28, and SETDB1 to deposit and maintain H3K9me3 at the 3' exons of zinc-finger genes. Related references: PMID:27029610
  • Acts as a negative regulator of histone macroH2A1 incorporation at telomeres and the alpha-globin cluster.
  • Participates in allele-specific gene expression at the imprinted IGF2/H19 locus, facilitating chromatin occupancy of SMC1 and CTCF on the maternal allele.
  • Subcellular localization: predominantly nuclear, associated with chromosomes, telomeres, and PML bodies.
  • Tissue specificity: ubiquitously expressed in human tissues.

Experimental Guidance and Technical Tips

  • The immunogen used is a synthetic peptide corresponding to amino acids 2100–2200 of human ATRX. Consider verifying antibody specificity using peptide competition assays or by comparing signals in ATRX knockout versus wild-type samples.
  • For Western blotting, note that ATRX is a large protein (~283 kDa); use a low-percentage polyacrylamide gel (e.g., 6–8%) and ensure efficient transfer by extending transfer times or using optimized protocols for high-molecular-weight proteins.
  • This antibody detects both human and mouse ATRX; if working with mouse tissues, validate cross-reactivity in your model system.
  • In ELISA applications, the immunogen peptide can be employed as a coating antigen to determine antibody titer or to set up competitive assays.
