Biological Background: ATPIF1 Function and Localization

  • ATPIF1 is an endogenous inhibitor of F(1)F(o)-ATP synthase that limits ATP depletion when mitochondrial membrane potential falls below a threshold, preventing the enzyme from acting as an ATP hydrolase (PubMed:35926043). Related references: PMID:35926043
  • Indirectly regulates heme synthesis in erythroid tissues by modulating mitochondrial pH and redox potential, which facilitates ferrochelatase (FECH)-mediated iron insertion into protoporphyrin IX.
  • Exclusively localized to the mitochondrion, with a nuclear-encoded precursor containing a cleavable transit peptide.
  • Features a coiled-coil domain, undergoes alternative splicing, and is regulated by phosphorylation.
  • The mature protein has a predicted molecular weight of approximately 12.2 kDa (UniProt Q9UII2), consistent with the ~12 kDa observed band.
