Biological Background: ATPB Function and Localization

  • ATP5B (ATP synthase subunit beta) is the catalytic beta subunit of mitochondrial F1F0 ATP synthase (Complex V), driving ATP synthesis from ADP using the proton gradient across the inner membrane. Related references: PMID:37244256

  • The enzyme consists of a soluble F1 catalytic head and a membrane-bound F0 proton channel, connected by a rotating central stalk and a stationary peripheral stalk. Related references: PMID:37244256

  • Together with the alpha subunit (ATP5F1A), ATP5B forms the catalytic core of the F1 domain. Related references: PMID:37244256

  • Localized to the mitochondrial inner membrane, where it functions as part of the oxidative phosphorylation machinery.

  • Subject to acetylation and phosphorylation; classified as a glycoprotein.

  • Mutations in ATP5B have been associated with dystonia and other mitochondrial disorders.

  • Possesses nucleotide-binding and magnesium-binding sites essential for its catalytic activity.

  • The ATP5F1B gene is also known as ATP5B, ATPMB, ATPSB, and HEL-S-271.

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 230–529, a region likely conserved across species, supporting the observed cross-reactivity with mouse and rat.

  • For western blotting, the predicted molecular weight is ~57 kDa; use appropriate gel percentage and transfer conditions for proteins of this size.

  • For IHC-P and IF/ICC, optimize fixation and permeabilization protocols to ensure accessibility of mitochondrial membrane antigens.

  • The antibody is compatible with ELISA; consider using recombinant ATPB protein as a coating antigen when developing quantitative assays.
