Biological Background: ATP6V1E1 Function and Localization
- ATP6V1E1 encodes the V-type proton ATPase subunit E 1, also known as V-ATPase 31 kDa subunit (p31), Vacuolar proton pump subunit E 1, and ATP6E/ATP6E2. Alternative splicing may generate multiple isoforms.
- As a subunit of the peripheral V1 complex of V-ATPase, it participates in ATP hydrolysis to drive proton translocation across membranes, thereby acidifying intracellular compartments such as lysosomes, endosomes, and secretory vesicles (PubMed:32001091, PubMed:33065002). Related references: PMID:32001091 PMID:33065002
- V-ATPase also localizes to the plasma membrane in certain cell types, contributing to extracellular acidification and pH regulation (PubMed:32001091). Related references: PMID:32001091
- The V-ATPase complex acts as an mTORC1 activator on lysosomal membranes by facilitating the guanine nucleotide exchange factor (GEF) activity of the Ragulator complex, thus promoting mTORC1 recruitment and anabolic signaling (PubMed:22053050, PubMed:39695235). Related references: PMID:22053050 PMID:39695235
- Subcellularly, ATP6V1E1 is found on the apical cell membrane, cytoplasmic vesicle membranes (synaptic vesicles, clathrin-coated vesicles), and lysosome membrane.
- The protein is ubiquitously expressed, with high levels in kidney (particularly distal nephron), skin, and widespread detection across tissues (PubMed:29993276, PubMed:12036578, PubMed:28065471). Related references: PMID:29993276 PMID:12036578 PMID:28065471
- Post-translational modifications include acetylation and phosphorylation, as indicated by proteomics studies, which may regulate its assembly or activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 77-226 of human ATP6V1E1, a region that is well-conserved across species, supporting the observed cross-reactivity with mouse and rat.
- For Western blotting, a band of approximately 26kDa is expected. Use appropriate positive controls such as HeLa or kidney tissue lysates, and consider including a negative control.
- In immunofluorescence/ICC, the antibody should recognize V-ATPase in vesicular structures and possibly at the plasma membrane. Optimization of permeabilization and fixation conditions is recommended for each cell type.
- ELISA applications may utilize the recombinant immunogen or peptide fragments as coating antigens; pre-validation of the antibody in the chosen assay format is advised.
- As a polyclonal antibody, it may detect multiple isoforms arising from alternative splicing; this property can be advantageous for detecting total ATP6V1E1 levels.
- Although cross-reactivity with mouse and rat is indicated by the supplier, individual validation in your specific sample system is recommended due to potential species-specific variations in antigen accessibility.
