Biological Background: ATP6V1B1 Function and Localization
- ATP6V1B1 encodes the kidney isoform of the V-ATPase B subunit, also known as the endomembrane proton pump 58 kDa subunit, vacuolar proton pump subunit B1, and previously designated as ATP6B1, VATB, or VPP3.
- As a non-catalytic subunit of the V1 peripheral complex, it is essential for the assembly and activity of the vacuolar-type H+ ATPase, which acidifies intracellular compartments and the extracellular environment (PubMed:16769747, PubMed:32001091). Related references: PMID:16769747 PMID:32001091
- In renal intercalated cells, ATP6V1B1 mediates proton secretion into urine, ensuring correct urinary acidification (PubMed:16769747). Related references: PMID:16769747
- Required for optimal olfactory function by mediating acidification of the nasal olfactory epithelium (By similarity).
- The protein localizes to apical and basolateral cell membranes, consistent with its role in polarized proton transport.
- Primarily expressed in kidney, specifically in the early distal nephron including thick ascending limbs and distal convoluted tubules, and also found in the cochlea and endolymphatic sac (PubMed:16769747, PubMed:29993276, PubMed:9916796). Related references: PMID:16769747 PMID:29993276 PMID:9916796
- Mutations in ATP6V1B1 are associated with sensorineural deafness and distal renal tubular acidosis (DRTA2), highlighting its critical role in hearing and acid–base homeostasis.
- Keywords: ATP-binding, nucleotide-binding, hydrogen ion transport, ion transport, membrane protein – these reflect its core biochemical functions.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal region (aa 1–280) of human ATP6V1B1; verify that the antibody recognizes full-length endogenous protein in your sample system.
- For Western blot, consider loading 20–30 μg of kidney tissue lysate (human, mouse, or rat) and expect a band near 57 kDa; confirm specificity using a positive control such as transfected cell lysate.
- In immunofluorescence (IF-P), validate staining patterns in kidney sections or cultured renal epithelial cells; apical and basolateral membrane localization should be expected.
- For ELISA applications, optimize coating antigen and detection conditions using recombinant protein or cell lysates; this antibody may capture or detect ATP6V1B1 in sandwich ELISA setups.
- When using mouse or rat samples, cross-reactivity has been confirmed, but always include appropriate species-specific secondary antibodies and controls.
