Biological Background: ATP5L Function and Localization
- ATP5L, also known as ATP5MG or ATP synthase membrane subunit g, is a subunit of the mitochondrial F(0) membrane domain of ATP synthase (Complex V). Related references: PMID:37244256
- The ATP synthase complex harnesses the proton gradient across the inner mitochondrial membrane to drive ATP production from ADP, with the F(0) domain forming the proton channel.
- Subunit g, along with other F(0) components, is essential for the structural integrity and proton translocation activity of Complex V.
- Under physiological conditions, the enzyme functions exclusively as an ATP synthase; artificial conditions in vitro can activate its reverse ATP hydrolase activity.
- The protein localizes to the mitochondrion inner membrane.
- Key post-translational modifications include acetylation, which may regulate enzymatic function or stability.
Experimental Guidance and Technical Tips
- For Western blot (WB), the antibody is expected to detect a band at approximately 11 kDa corresponding to endogenous ATP5L. Validate in your sample system and consider using mitochondrial fractions for enrichment.
- In immunofluorescence/immunocytochemistry (IF/ICC), mitochondrial staining should be observed. Co-localization with known mitochondrial markers is advised to confirm specificity.
- For ELISA, the antibody can be used to detect recombinant or purified ATP5L protein. Optimize coating and detection conditions.
