Biological Background: ATP5C1 Function and Localization
- ATP5C1 (ATP synthase F1 subunit gamma) is a critical subunit of the mitochondrial F1F0 ATP synthase (Complex V), the enzyme responsible for the synthesis of ATP from ADP and inorganic phosphate using the proton-motive force across the inner mitochondrial membrane. - This protein exists as two major isoforms: Heart isoform, predominantly expressed in cardiac and skeletal muscle, and Liver isoform, expressed in brain, liver, kidney, skin, intestine, stomach, and aorta, reflecting tissue-specific energy demands.
- As the gamma subunit of the central stalk, ATP5C1 couples proton flow-driven rotation in the F0 membrane domain to conformational changes in the F1 catalytic head, enabling ATP synthesis.
