Biological Background: ATP5A1 Function and Localization

  • ATP5A1 encodes the alpha subunit of mitochondrial ATP synthase (Complex V), which synthesizes ATP from ADP using the proton gradient generated by the electron transport chain. Related references: PMID:37244256
  • Together with the catalytic subunit beta (ATP5F1B), it forms the catalytic core of the F1 domain; however, subunit alpha does not bear the catalytic high-affinity ATP-binding sites. Related references: PMID:37244256
  • Binds the bacterial siderophore enterobactin and can promote mitochondrial accumulation of enterobactin-derived iron ions. Related references: PMID:30146159
  • Located primarily in the mitochondrial inner membrane, but also reported at the cell membrane.
  • Expressed in fetal lung, heart, liver, gut, and kidney, with higher levels in fetal brain, retina, and spinal cord.
  • Undergoes post-translational modifications including acetylation, glycation, methylation, and phosphorylation.
  • Associated with primary mitochondrial disease; mutations have been linked to disorders such as COXPD22.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a central region (aa 200–300) of human ATP5A1, which is highly conserved, likely contributing to the observed cross-reactivity with mouse and rat.
  • For Western blotting, a band at approximately 60 kDa is expected; consider using appropriate loading controls and pre-stained markers.
  • In immunofluorescence and immunohistochemistry, validate staining patterns in your specific tissue or cell model, as expression can vary. Mitochondrial localization may require counterstaining with organelle markers.
  • For immunoprecipitation, titrate the antibody amount to optimize target capture while minimizing background.
  • Because ATP5A1 is a mitochondrial protein, sample preparation strategies such as subcellular fractionation may improve detection sensitivity in some applications.
