Biological Background: ATOH8 Function and Localization

  • ATOH8 is a basic helix-loop-helix (bHLH) transcription factor, also referred to as bHLHa21 or hATH6.
  • It binds to the canonical E-box DNA sequence 5'-CANNTG-3', possibly as a heterodimer with other bHLH proteins, to regulate gene transcription. Related references: PMID:24236640
  • ATOH8 regulates endothelial cell proliferation, migration, and tube-like structure formation, and modulates endothelial cell differentiation through NOS3. Related references: PMID:24463812
  • It may participate in neuronal cell lineage specification in the brain, kidney development, and podocyte differentiation, and modulates NEUROG3-initiated differentiation programs.
  • During myogenesis, ATOH8 may play a role in the transition from myoblast proliferation to differentiation.
  • It positively regulates HAMP transcription by direct promoter binding and by enhancing SMAD phosphorylation. Related references: PMID:24236640
  • ATOH8 represses NEUROG3-dependent gene activation through heterodimerization with general partners and potentially through direct DNA binding.
  • Subcellular localization includes the nucleus (particularly nuclear speckles) and cytoplasm.
  • The protein is expressed in lung, liver, kidney, heart, pancreas, and endothelial cells of umbilical vessels.
  • Key functional annotations include alternative splicing, developmental protein, differentiation, DNA-binding, neurogenesis, and transcription regulation.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide from the N‑terminal region (amino acids 1–100). For Western blotting, consider optimizing blocking and washing conditions to reduce non‑specific binding typical of polyclonal antibodies.
  • In ELISA applications, use appropriate coating buffers and validate the antibody against the specific peptide or recombinant protein to ensure reactivity.
  • The predicted molecular weight is 35 kDa, but actual migration in SDS‑PAGE may vary due to post‑translational modifications; always run appropriate molecular weight markers and positive controls.
  • For indirect detection, choose a secondary antibody that matches the rabbit host and is validated for your detection method, and include proper control experiments.
